Abstract:

Genetically engineered immune cells such as T cells capable of secreting an interleukin-12 protein, for example, upon activation of the T cells. Such genetically engineered immune cells may further express a chimeric antigen receptor (CAR) targeting an antigen of interest, e.g., a tumor-associated antigen, a disrupted T cell receptor alpha chain constant (TRAC) gene, a disrupted beta-2-microglubulin (.beta.2M) gene, a disrupted gene encoding the antigen of interest, or a combination thereof.
