METHODS AND COMPOSITIONS FOR MESSENGER RNA PURIFICATION

Abstract:

The present invention provides, among other things, methods for purifying high quality messenger (mRNA) suitable for clinical use. The present invention is, in part, based on surprising discovery that capping and tailing mRNA in reaction buffer having a pH lower than 8.0 and MgCl.sub.2 at a concentration of less than 1.25 mM can increase RNA integrity of final mRNA product. Thus, the present invention provides an effective, reliable, and efficient method of manufacturing high quality RNA at large scale for therapeutic use.
